Radiolabeling of Red Blood Cells with 51Cr Using Citrate Washing Procedure

1 | 10 mL of blood are placed in sterile, empty 25-mL vial |
2 | 2 mL of acid-citrate-dextrose anticoagulant are added and swirled gently to mix |
3 | 5 mL of citrate-phosphate buffer (pH 6.4–6.9) are added and swirled gently to mix |
4 | Cell suspension is centrifuged gently, and supernatant is removed/discarded |
5 | Washed cells are resuspended using normal saline solution |
6 | 0.925 MBq (25 μCi) of 51Cr sodium chromate is added and swirled gently to mix |
7 | Incubation is performed at room temperature with occasional mixing for 20 min |
8 | Cell suspension is centrifuged gently, and supernatant is removed/discarded |
9 | 51Cr-labeled cells are resuspended using normal saline solution |
